Dietary UMP Shows Brain Cholinergic‑Modulating Potential for Age‑Related Cognitive Decline Research

An animal study published in Brain Research demonstrates that dietary supplementation with uridine‑5′‑monophosphate (UMP) raises striatal phospholipid levels and boosts the content and release of acetylcholine, offering new insights for interventions against age‑related cognitive decline. Acetylcholine is a vital brain neurotransmitter, and its functional decline is closely linked to age‑related deterioration in memory and learning capacity.

 

UMP serves as a precursor for membrane phosphatidylcholine. In animal tests, aged rats fed UMP‑supplemented diets for one or six weeks exhibited markedly elevated basal extracellular acetylcholine levels in the striatum. Low‑dose UMP treatment for one week increased total brain‑tissue acetylcholine content by 16%. Upon atropine stimulation, the UMP group showed far greater acetylcholine release compared with controls. Similar effects were also observed in young rats.

 

Experiments confirm UMP does not inhibit acetylcholinesterase activity. It works by stimulating the synthesis of brain membrane phospholipids including phosphatidylcholine to increase choline supply for acetylcholine production. It also supports the formation of synaptic membranes and vesicles, further facilitating cholinergic neurotransmission.

 

Researchers note orally administered UMP is metabolized into uridine which crosses the blood‑brain barrier. The findings suggest UMP holds promise as a nutritional or drug candidate to improve cholinergic signaling. Nevertheless, results are limited to rat studies; further pre‑clinical and human trials are required to evaluate its real‑world value for age‑related cognitive impairment.
